A Cipk23 Interacting Transporter Protein is Implicated in Cd Tolerance and Phytoremediation Through Vesicle-Dependent Compartmentalization in Roots
Cadmium (Cd) pollution in environment imposes serious risk to living organisms, and uptake of Cd by crops results in chronic dietary exposure. Therefore, it is urgent to unravel mechanisms by which plants tolerate Cd and find ways to remediate Cd-polluted soils. Here, we identified a transporter protein by screening interaction partners with a well-known protein kinase, CIPK23. Loss-of-function mutation of CIPK23 displayed increased sensitivity to Cd compared to wild-type plants. A CIPK23 interacting transporter protein, CIP1, was screened by yeast two-hybrid screening assays and loss-of-function mutation of CIP1 showed decreased tolerance to Cd stress. CIP1 is localized to vesicles and has Cd transport activity. The expression of CIP1 was specifically induced by Cd and was mainly expressed at root epidermis. Furthermore, transgenic tobacco lines overexpressing CIP1 showed increased tolerance to Cd and had improved capability to accumulate Cd. However, the accumulation of Cd was confined to roots, suggesting that CIP1 could be an excellent transporter protein employed to cleanup Cd-polluted soils without the penalty of economic loss. Taken together, we identified and characterized a novel Cd transporter that has potentials for improving plant Cd tolerance and remediating Cd pollution in environment
