Melatonin Delays Leaf Senescence and Improvs Yield of Cucumber by Modulating Chlorophyll Degradation and Photosynthesis Inhibition of Psii and Psi
Decrease of chlorophyll and photosynthetic efficiency co-occur during aging-leaf senescence. Here, we reported the positive functions of melatonin (MT) on delaying leaf senescence of cucumber plants. Results showed that natural leaf senescence induced the accumulation of endogenous MT and spraying MT delayed leaf senescence depended on a concentration-dependent manner. Moreover, application of MT and transient over-expression of CsASMT , the key gene of MT synthesis, both suppressed leaf senescence simulated by darkness, as evidenced by decrease of senescence-related gene SAG12 and cell death-related gene PDCD expression, increase of chlorophyll content in terms of down-regulation of chlorophyll- degradation genes and normal chloroplast structure, as well as higher activity of PSI, PSII reaction center and electron transfer efficiency and photosynthesis -associated protein level, compared with the H 2 O-treated seedlings. Meanwhile, the data from field spraying experiments proved that MT indeed alleviated the decline of chlorophyll content and increased the photosynthetic efficiency in both functional and senescent leaves, which finally promoted the growth and yield of cucumber in solar-greenhouse. Together, these results implied that MT acts as a positive regulator to down-regulate the mRNA abundance of SAG12 and PDCD and alleviate the damage to photosynthetic apparatus via suppressing chloroplast disintegration and photosynthesis -associated protein degradation during leaf senescence
